For example,
I go to http://6abc.com/shots-fired-at-police-in-gloucester-twp-standoff-schools-closed/2927128/ and an ad is displayed before the actual video begins.

The video from comment 1 doesn't seem to have an ad anymore. Albeit other videos on the site do.
Update: it is just that you don't get ads all the time.

You need the following blocking filter
||content.uplynk.com/preplay/*
AND
and url rewrite as issue #6592 mentions.

- Resolution set to fixed
- Status changed from new to closed
With these two filters:
||content.uplynk.com/api/v3/preplay/* /(content.uplynk.com\/ext\/.*\.m3u8)\?.*$/$rewrite=$1
and the 3.1.0.2053 development build, it should work.

Seems to work in Chrome but in Firefox 60.0.1 the player is black. Tested with the suggested filters and ABP 3.1.0.2053

With these two filters
One filter is enough if you rewrite the parameter "ad" to something else.
example &ad=$rewrite=&abp=
